近日,山東銀豐生命科學研究院(銀豐生科院)所屬再生醫學研究所曲廷瑜教授帶領的研發團隊在線發表了題為“Mesenchymal stem cells from human umbilical cord regulate the expression of major histocompatibility complex in human neural stem cells and their lineages”的論(lun)文。

研(yan)究工作(zuo)由(you)銀豐生科(ke)院與山東大學齊(qi)魯(lu)醫(yi)院和首都醫(yi)科(ke)大學天壇醫(yi)院共同完成,將于2020年11月1日國際SCI雜志《Neuroscience Letters》刊出。

眾所(suo)(suo)周知(zhi),異體(ti)干(gan)(gan)細(xi)(xi)胞(bao)(bao)移植會(hui)引起(qi)機體(ti)的免(mian)疫排斥反應,而(er)導致免(mian)疫排斥的主要原(yuan)因與供體(ti)細(xi)(xi)胞(bao)(bao)攜帶的組織相容性(xing)復(fu)合體(ti) (Major Histocompability Complex:MHC) 抗(kang)原(yuan)表達有關。該(gai)團隊利用細(xi)(xi)胞(bao)(bao)共培(pei)養體(ti)系研究(jiu)了臍(qi)帶間充質干(gan)(gan)細(xi)(xi)胞(bao)(bao)(hUC-MSCs) 對人源性(xing)神(shen)經(jing)干(gan)(gan)細(xi)(xi)胞(bao)(bao)(hNSCs)及其所(suo)(suo)分化的各(ge)種(zhong)神(shen)經(jing)細(xi)(xi)胞(bao)(bao)MHC表達的免(mian)疫調控作用,發現(xian)在炎性(xing)狀(zhuang)態下,神(shen)經(jing)干(gan)(gan)細(xi)(xi)胞(bao)(bao)和(he)神(shen)經(jing)膠質細(xi)(xi)胞(bao)(bao)可(ke)表達大(da)量的MHC-II類抗(kang)原(yuan)分子(zi)。間充質干(gan)(gan)細(xi)(xi)胞(bao)(bao)可(ke)通(tong)過其免(mian)疫調節特性(xing)抑制神(shen)經(jing)干(gan)(gan)細(xi)(xi)胞(bao)(bao)和(he)神(shen)經(jing)膠質細(xi)(xi)胞(bao)(bao)MHC-II類抗(kang)原(yuan)分子(zi)的過度表達,從(cong)而(er)能避免(mian)機體(ti)免(mian)疫系統(tong)的激活而(er)降低(di)免(mian)疫排斥。

曲廷瑜(yu)教(jiao)授(shou)表(biao)示(shi),該研究從免(mian)(mian)疫(yi)學的(de)(de)角度闡明了(le)間充質干細(xi)胞(bao)(bao)對神經(jing)干細(xi)胞(bao)(bao)免(mian)(mian)疫(yi)原性(xing)(xing)的(de)(de)調節機(ji)制,探討(tao)了(le)兩種干細(xi)胞(bao)(bao)臨(lin)床配伍(wu)的(de)(de)協(xie)同(tong)性(xing)(xing);而最近與山東大學齊(qi)魯醫院陳允震教(jiao)授(shou)合作發表(biao)的(de)(de)神經(jing)干細(xi)胞(bao)(bao)和(he)間充質干細(xi)胞(bao)(bao)共同(tong)移植治(zhi)療脊髓損傷的(de)(de)研究《Cell Transplantation》則對這兩種干細(xi)胞(bao)(bao)聯合應(ying)用的(de)(de)可(ke)行性(xing)(xing)提供(gong)了(le)可(ke)靠(kao)的(de)(de)臨(lin)床前實驗依據。

目前臨(lin)床(chuang)上對(dui)眾多神(shen)(shen)(shen)經(jing)(jing)(jing)系統疾(ji)病治(zhi)療仍然(ran)束手無策,而神(shen)(shen)(shen)經(jing)(jing)(jing)干(gan)細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)移(yi)植(zhi)有望成(cheng)為這些疾(ji)病的(de)(de)(de)有效或治(zhi)愈的(de)(de)(de)手段(duan)。近年來,在(zai)銀豐生(sheng)科院曲(qu)廷瑜教(jiao)授(shou)團隊對(dui)神(shen)(shen)(shen)經(jing)(jing)(jing)干(gan)細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)的(de)(de)(de)應(ying)用開(kai)展(zhan)了(le)系列性的(de)(de)(de)研(yan)究工(gong)作,相(xiang)關研(yan)究成(cheng)果(guo)分別發表在(zai)《Stem Cells International》,《Frontiers Neural Circuits》,《Neuroscience Letters》,《NeuroReport》,和(he)《中(zhong)華細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)與干(gan)細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)雜志》等(deng)雜志,內容(rong)包(bao)括了(le)神(shen)(shen)(shen)經(jing)(jing)(jing)干(gan)細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)制備(bei),神(shen)(shen)(shen)經(jing)(jing)(jing)細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)的(de)(de)(de)誘導分化(hua)(hua),功能性多巴胺神(shen)(shen)(shen)經(jing)(jing)(jing)元的(de)(de)(de)獲(huo)得(de)和(he)優化(hua)(hua),自體嗅鞘神(shen)(shen)(shen)經(jing)(jing)(jing)細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)分離(li)和(he)培養,以及腦缺血(xue)動物模型的(de)(de)(de)建立和(he)神(shen)(shen)(shen)經(jing)(jing)(jing)干(gan)細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)條(tiao)件培養液對(dui)腦缺血(xue)再灌流損傷的(de)(de)(de)治(zhi)療等(deng)。這些研(yan)發成(cheng)果(guo)進(jin)一步推動了(le)神(shen)(shen)(shen)經(jing)(jing)(jing)干(gan)細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)的(de)(de)(de)臨(lin)床(chuang)轉化(hua)(hua),對(dui)銀豐生(sheng)物集團即(ji)將(jiang)開(kai)展(zhan)的(de)(de)(de)神(shen)(shen)(shen)經(jing)(jing)(jing)干(gan)細(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)臨(lin)床(chuang)試驗和(he)藥(yao)品申(shen)報工(gong)作奠(dian)定(ding)了(le)基礎。
